Scientists Develop RK-251 Cancer Drug
Indian researchers announced RK-251 on 19 August 2026 as a smart cancer drug that remains inactive in healthy tissue and activates inside tumour cells. The drug was developed through a collaboration led by Dr Asis Bala of the Institute of Advanced Study in Science and Technology and Dr K.P. Bhabak of the Indian Institute of Technology Guwahati.
Smart Drug Design
RK-251 is designed as a prodrug, which is a compound that becomes active only after a chemical trigger. In RK-251, the trigger is reactive oxygen species, or ROS, which are present at higher levels in many cancer cells than in normal cells.
When ROS activates RK-251 inside the tumour microenvironment, the drug releases NBDHEX, an anticancer compound that blocks proteins used by cancer cells for survival. This design aims to limit drug action to cancer cells and reduce exposure of healthy cells to the active compound.
Preclinical Testing
Preclinical laboratory studies tested RK-251 against aggressive triple-negative breast cancer cells, a subtype of breast cancer that lacks oestrogen receptor, progesterone receptor, and HER2 expression. The drug showed targeted activity against these cells while sparing healthy cells in laboratory conditions.
Toxicity and behavioural studies in zebrafish embryos, scientifically known as Danio rerio, found no obvious signs of toxicity. Fluorescence-based tests also confirmed activation of the drug in high-ROS environments.
