CLOVES Syndrome Awareness Day is observed every year on 3rd August to support affected individuals and promote early diagnosis and treatment. CLOVES stands for Congenital Lipomatous Overgrowth, Vascular malformations, Epidermal nevi, and Spinal/Skeletal anomalies. CLOVES Syndrome is a rare overgrowth genetic disorder caused by mutations in the PIK3CA gene. The PIK3CA gene affects the PI3K enzyme, which regulates cell functions and may cause cancer if mutated. It belongs to a group of disorders called PIK3CA-related overgrowth spectrum (PROS). The syndrome was formally classified in 2007–2009 but may have been first documented in 1867.
